It is activated by a set of transfected plasmids, and is inhibited by a cell line producing v-erb-a protein. We have used these features to define the phenotype of v-erb-b induced transformation. In transfection experiments, v-erb-b transformed cells could be divided into a group that could be inhibited by v-erb-a (12%, n = 62) and one that was insensitive (88%, n = 78). The sensitive cells had a lower saturation density than the insensitive ones (1.03 +/- 0.06 vs 1.16 +/- 0.12), and a higher plating efficiency (24 +/- 1% vs 13 +/- 2%). These two properties could be corrected by addition of 1% serum (saturation density, 1.11 +/- 0.07; plating efficiency, 13 +/- 2%) or by exposure of transfectants to 3% oxygen (saturation density, 1.05 +/- 0.05; plating efficiency, 17 +/- 1%). Both abilities were more marked in cells transformed in the presence of both v-erb-a and v-erb-b. An elevation in the level of c-Ha-ras and t-Ha-ras oncogene transcripts was detected by Northern blotting in sensitive cells and was down-regulated in insensitive cells after serum or oxygen treatments. The phenotypes of the latter were characterized by a more pronounced alteration of their morphology, growth kinetics, plating efficiency, saturation density, and specific binding of epidermal growth factor (EGF) than was the case with the sensitive cells.
