Their biological functions include structural stabilization of glycoproteins, intracellular biosynthesis, and degradation of beta-D-galactosyl oligosaccharides and oligosaccharide intermediates derived from N-linked glycoproteins. In this study, a new gradient of gradient-elution HPLC was developed for the determination of beta-galactosidase activity. Oligosaccharides that consisted of one to seven beta-linked D-galactose residues were separated in the gradient-elution mode by using the TSK G3000 SWXL column (Tosoh, Tokyo, Japan) and applied to the simultaneous determination of three classes of bacterial beta-galactosidases. The developed method resolved the individual species of the beta-linked D-galactose oligosaccharides from natural glycoproteins, beta-galactosidases, and synthetic substrates. Using the stepwise gradient of hexane and an acetonitrile/50 mM sodium citrate (pH 5.0) eluent, the separation of natural oligosaccharides and synthetic substrates was also possible. Gradient-elution HPLC of a mixture of disaccharides obtained from natural glycoproteins with beta-galactosidase of Pseudomonas aeruginosa produced results similar to those from the simultaneous determination of beta-galactosidases. The detection limit of this HPLC method for the determination of beta-galactosidase activity was 25 mU/ml, which was a similar result to that of the present standard spectrophotometric assay.
