Loss of PPP4C results in serum-independent NF-kB activation and lymphadenopathy in the B-cell receptor?deficient mouse strain. The studies proposed herein are centered on the regulation of PLC?2 ubiquitination, a key event in lymphocyte activation. The first aim will determine the molecular mechanism by which PPP4C regulates PLC?2. Preliminary data indicate that PPP4C binds to PLC?2 directly and that the activity of PPP4C is regulated by dephosphorylation at Ser-173. The second aim will identify the site on PLC?2 that is responsible for deubiquitination and is a regulator of PLC?2 activity.
